My wife and I are planning our 3rd trip to Desire this Feb. We were wondering if its possible to choose your room # when booking directly through Desire.

If you use the Express Check form on the Desire site
http://desire.originalresorts.com/express_checkin.aspJust fill in your request for a particular room in the "Special Requirements" area of the form.
Desire offers no guarantees of anyone getting a specific room but they do try hard to accommodate your request
