Desire has an "Express Check-in" system that you can use to request specific rooms. The link URL is http://desire.originalresorts.com/express_checkin.asp. You can complete the express check-in form any time within 2 weeks of your arrival.

They won't guarantee you get the room you want, but they will TRY to give you the room you want. Your best bet is to give them a short, prioritized list of rooms you'd like so if your first choice isn't available, your second or third choice might be.
